QPCR Preparation

A tube or 96-well plate can be used for qPCR. Reactions take place in each well of the plate (or tube), which is loaded with target DNA, controls and standards. Analysis assumes an equal volume in each well containing:

  1. Template (DNA, cDNA, or RNA if one-step reverse trancription is done in the well)
  2. Primers (specific for the DNA target)
  3. qPCR mix
    1. Fluorescent dye
    2. DNA polymerase
    3. Deoxyribonucleotide triphospates (dNTPs)
    4. Buffers
  4. Water to specified volume
